Zusammenfassung
Am Modell einer menschlichen Lymphoblastenkultur wird demonstriert, daß die zytostatische Wirkung von Methotrexat® durch Methyl-Tetrahydrofolsäure aufgehoben werden kann. Zur Deutung der Ergebnisse wird folgende Hypothese diskutiert: Die Wirkung der 5-Methyl-Tetrahydrofolsäure und auch der klinisch als Methotrexat-Antidot angewendeten 5-Formyl-Tetrahydrofolsäure (Leucovorin®) beruht auf der Übertragung der präformierten bzw. der durch enzymatische Reduktion gebildeten Methyl-Gruppe auf Homozystein in der Vitamin-B12-abhängigen Methionin-Synthetase-Rekation. Dabei wird Tetrahydrofolsäure frei und steht für die Aktivierung und Übertragung von C1-Einheiten zur Verfügung. Die de-novo-Synthese von Purin- und Pyrimidinnukleotiden kann wieder ablaufen.
Summary
As demonstrated with the model system of cultured human lymphoblasts the cytostatic effect of amethopterine (Methotrexate®) can be overcome by methyl-tetrahydrofolic acid. This result is discussed as follows: the effect of 5-methyl tetrahydrofolic acid as well as 5-formyl tetrahydrofolic acid (Leucovorin®) which is clinically used as a potential Methotrexate-antidot depends on the transferation of the preformed or enzymatically produced methyl group to homocysteine. The vitamin B12-dependent methionine-synthetase is the key enzyme in this reaction: it liberates free tetrahydrofolic acid which is needed to activate C1-compounds and to transfer them into the de novo synthesis of purine and pyrimidine nucleotides.
