Zusammenfassung
Die verschiedenartige Wirkung von Glukose und Purinnucleosiden (Inosin) sowie Nucleosid-Purin-Gemisches (Inosin+Adenin) auf den Stoffwechsel frischer und gealterter Erythrozyten wurde untersucht. Als besondere Stoffwechselleistung wurde die Fähigkeit, Hb III zu Hb II zu reduzieren analysiert.
Es wurde festgestellt, daß Purinnucleoside bei 80 bis 100 Tage gelagerten Erythrozyten sehr gute Hb III-Reduktion bewirken, während Glukose unwirksam ist. Glukose kann jedoch dann wieder metabolisiert werden, wenn die gealterten Zellen mit Purinnucleosiden vorbehandelt werden.
Weder Hexokinase noch Phosphoglycerinaldehyddehydrogenase, noch Glukose-6-Phosphatdehydrogenase werden während der Lagerung inaktiv. Lediglich der während der Konservierung in ACD-Milieu absinkende ATP-Gehalt bewirkt, daß die Zellen keine Glukose mehr aufnehmen und in den anaeroben und aeroben Abbauweg einschleusen können.
Hb III kann von jodacetatvergifteten Erythrozyten nur dann zu Hb II reduziert werden, wenn wasserstoffübertragende Farbstoffe die Verbindung zwischen TPNH und Hb III herstellen. Die TPNH-abhängige Reduktase ist nur in Gegenwart solcher Farbstoffe wirksam.
Summary
The differing effects of glucose and purine nucleosides (Inosin), and also of nucleoside-purine (Inosin+Adenin) on the metabolism on fresh and aged erythrozytes has been investigated. The particular metabolic effect analysed was the power of reducing Hb III to Hb II.
It was found that purine nucleosides caused good Hb III reduction in erythrocytes stored for 80 to 100 days, whereas glucose was ineffective. Glucose can, however, again be metabolised if the aged erythrocytes are first treated with purine nucleosides.
During storage, phosphoglycerol-aldehyde-hydrogenase, hexokinase and glucose-6-phospate-hydrogenase do not become inactive. The fall in ATP content during storage in ACD mediums has the effect of preventing the cells from taking up any glucose and of enabling them to be diverted into the anaerobic and aerobic breakdown process.
Hb III can only be converted into H III in erythrocytes poisoned with iodine acetate if hydrogen-carrying pigments produce a compound of TPNH and Hb III. TPNH-dependent reductase is only active in the presence of such pigments.
