Elektronenmikroskopische Untersuchung über die Blutbilung in der fetalen Rattenleber (Tag 14 a.p.) nach Gabe von Chloramphenicol und Oxytetrazyklin

Zusammenfassung

Wir haben die Blutbildung in der fetalen Leber von Ratten vor und nach Gabe von Chloramphenicol (CAP) und Oxytetrazyklin (OTZ) am Tage 14 a.p. untersucht. Dabei fanden sich an den Vorstufen der roten Blutreihe charakteristische Veränderungen. Die Proerythroblasten und Erythroblasten zeigen eine Aufhellung des Zytoplasmas und eine Unregelmäßigkeit der Zellkonturen. Diese Elektronendurchlässigkeit kommt einerseits durch eine Abnahme der Polysomen, bei den polychromatophilen Eythroblasten aber auch durch einen Verlust des Hämoglobins im Zytoplasma zustande. Die Mitochondrien vieler Proerythroblasten und basophiler Erythroblasten sind zahlenmäßig vermindert und zeigen zwei unterschiedliche Schädigungsmuster:

    Eine Schwellung mit Auflösung der Cristae und Auftreten von tubulären und vesikulären Strukturen.

  2. 2.

    Myelinfiguren im Inneren der Mitochondrien.

Die Sidersomen im Zytoplasma sind nich vermehrt. Der Mechanismus, der nach Gabe von OTZ und CAP zu diesen Veränderungen führt, wird diskutiert.

Summary

We studied the erythropoiesis in the foetal liver of rats on day 14 a.p. without and after doses of CAP and OTC. Characteristic changes could be found in the precurors of the red cell series. The proerythroblasts and erythroblasts showed a lightening of the cytoplasma and an irregular cell contour. This increased electron transmission is a result of the decrease in polysomes and in the polychromatophle erythroblasts, also a result of the loss of haemoglobin in the cytoplasma. The mitochondria of many result of the loss of haemoglobin in the cytoplasma. The mitochondria of many proerythroblasts and basophilic erythroblasts are decreased in number and show two different types of damage:

  1. 1.

    A swelling accompanied with dissolution of the cristae and the appearance of tubular and vesicular structures.

  2. 2.

    Myelin figures inside the mitochondria.

There was no increase of the siderosomes in the cytoplasma. The mechanism which leads to these chantes after doses of OTC and CAP is discussed.
