Zusammenfassung
Biochemische Untersuchungen an Erythrozyten-Präparationen von unbehandelten und von mit Acetyl-Phenylhydrazin vorbehandelten Ratten haben gezeigt, daß der Retikulozyt der Ratte ein vollständiges adrenergisches β-Rezeptor-Effektor-System enthält. Im einzelnen wurden folgende Ergebnisse erhalten: 1. Die in Erythrozyten von Ratten nachweisbare Adenylzyklase-Aktivität ist fast ausschließlich in den Retikulozyten lokalisiert. 2. Die Aktivität der in retikulozytenreichen Präparationen von Erythrozytenschatten nachweisbaren Adenylzyklase ist spezifisch stimulierbar durch β-Sympathomimetika; diese Wirkung wird durch β-Sympatholytika spezifisch gehemmt. 3. Die Aktivität des zyklisches Adenosin-3′5′-monophosphat inaktivierenden Enzyms ist in retikulozytenreichen Präparationen signifikant höher als in Präparationen aus retikulozytenarmem Blut. 4. In Erythrozytenschatten kann eine durch zyklisches Adenosin-3′5′-monophosphat stimulierbare Protein-Phosphokinase-Aktivität nachgewiesen werden, die in retikulozytenreichen Präparationen wiederum wesentlich höher ist. 5. In Erythrozytenschatten der Ratte sind Brenzkatechinamin-inaktivierende Enzyme nachweisbar: In retikulozytenreichen Präparationen wird — im Gegensatz zu Präparationen aus normalem Blut — Monoaminoxydase-Aktivität gefunden; die Aktivität der Catechol-O-methyltransferase ist in retikulozytenreichen Präparationen mehrfach höher.
Summary
Biochemical investigations in erythrocyte preparations from untreated and from acetyl-phenylhydrazine-prettreated rats have shown that rat reticulocytes contain a complete β-adrenergic receptor-effector system. The following experimental results were obtained: 1) Adenyl cyclase activity in rat erythrocytes is nearly exclusively localized in the reticulocytes. 2) Adenyl cyclase activity in ghost preparations from reticulocyte-rich blood of rats (produced by pretreating the animals with acetylphenylhydrazine) is specifically stimulated by β-sympathomimetic agents; this stimulatory effect is specifically depressed by β-adrenergic blocking drugs. 3) Cyclic AMP inactivating phosphodiesterases in both membrane and cytoplasmic fractions show significantly higher activity in reticulocyte-rich than in reticulocyte-poor preparations. 4) The activity of a cyclic AMP-stimulated protein kinase is also markedly higher in ghost preparations from reticulocyte-rich blood. 5) Catecholamine-inactivating enzymes are present in membrane fractions from rat erythrocytes: Catechol-O-methyl-transferase activity is several-fold higher in preparations from reticulocyte-rich than in preparations from reticulocyte-poor blood; significant monoamine oxidase activity is only found in reticulocyte-rich preparations.
