Differenzierung von Plaquevarianten des Herpes-simplex-Virus durch Gradientenzentrifugation und Säulenchromatographie

Zusammenfassung

Die beiden Herpes-simplex-Varianten fR- und rR-Typ konnten durch physikalische Methoden differenziert werden. Im Caesiumchloridgradienten weisen sie eine Dichtedifferenz von 0,6% auf.

Bei der Chromatographie auf ECTEOLA-Säulen ist auch ein unterschiedliches Elutionsverhalten beobachtet worden. Dieses Verfahren erlaubt ferner eine weitgehende Elimination von Fremdprotein und somit eine Reinigung des Virus.

Summary

The two variants of the “JES” herpes simplex strain causing the plaque types fR and rR respectively have been differentiated by physical methods. In a cesium chloride density gradient the two variants show a density difference of 0.6%. On chromatography in ECTEOLA columns the elution behaviour of the two variants is different. This chromatography constitutes a purification procedure as foreign protein is eliminated to a considerable extent.
