Expression ofDrosophila hsp 70-CAT hybrid gene inAedes cells induced by heat shock

Abstract

DNA-mediated transfer of a Drosophila hsp 70-CAThybrid gene into DrosophilaS3 cells leads to the appearance of heat shock (37° C)-inducible chloramphenicol acetyltransferase (CAT) activity. When this hybrid gene construction was transfected into cultured Aedes, Plodia,or Manducacells, only trace levels of heat-inducible CAT activity were observed. Induction could be somewhat improved by using Schneider's Drosophilamedium for transfection. In the case of Aedescells, levels of CA T induction comparable to that seen using Drosophilacells could be achieved by raising the heat-shock temperature to 41° C, a treatment which is lethal to Drosophila.
