Zusammenfassung
Bei Patienten mit einer obliterierenden Arteriosklerose (Verschlüsse im Bereich der unteren Extremitäten und der Coronararterien) fand sich eine gegenüber Normalpersonen signifikant gesteigerte Thrombocytenadhäsivität sowie ein ebenfalls signifikant erniedrigter intracellulärer Kaliumgehalt der Thrombocyten. Durch Aufbewahrung des plättchenreichen Plasmas bei Zimmertemperatur über 4 Std konnte auch bei normalen Thrombocyten eine Steigerung ihrer Adhäsivität und gleichzeitig eine Abnahme ihres Kaliumgehaltes hervorgerufen werden. Kaliumverlust und Zunahme der Plättchenklebrigkeit werden als Zeichen einer Frühschädigung der Thrombocyten aufgefaßt und eine erhöhte Permeabilität der Thrombocytenmembran als Ursache beider Veränderungen diskutiert. Die Bedeutung der Befunde im Rahmen von thrombotischen Komplikationen einer Arteriosklerose wird besprochen. Die Ursache einer Thrombocytenmembranschädigung in vivo ist unklar.
Summary
Platelets of patients whith obstruction of the leg arteries and coronaries (obliterating arteriosklerosis) show a significantly increased adhesiveness as compared to those of normal individuals. Furthermore, the intracellular potassium content of the platelets is in these cases greatly reduced. When plasma enriched in platelets was kept for about four hours at room temperature, even normal platelets showed increased adhesiveness and at the same time a reduction in potassium content. The reduced potassium content and increased adhesiveness of the platelets are interpretet as early signs of damage. The author discusses an increased permeability being the cause for both phenomena. The value of these findings in connection with thrombosis occuring in the course of arteriosclerotic disease is discussed. The cause of the membrane damage of the platelets in vivo remains unknown.
