The polypeptide chain structure of immunoglobulins. III. experimental studies on the recombination of labelled myeloma polypeptide chains

Summary

The extent of the recombination of heavy and light polypeptide chains of myeloma proteins has been measured by gelfiltration experiments after labelling of individual myeloma proteins by131I and125I. The recovery of 7s-molecules was calculated from the percentage of light chains diverted by heavy chains from the original position in the elution diagram. Recombination of heterologous polypeptide chains at a rate of 93% was observed. Immunoelectrophoretically, this “hybridized” immunoglobulin molecule showed the characteristics of an IgG molecule. The results confirm, that the experimental procedure, used for recombination experiments with several invidual myeloma proteins can be applied to labelled proteins as well.

Zusammenfassung

Das Ausmaß der Rekombination leichter und schwerer Polypeptidketten von Myelom-Proteinen wurde durch Gelfiltration nach Markierung individueller Myelom-Proteine durch131J und125J gemessen. Die Ausbeute an 7s-Molekülen wurde aufgrund des Prozentsatzes der leichten Ketten berechnet, die durch die schweren Ketten aus ihrer ursprünglichen Position im Elutionsdiagramm abgelenkt wurden. Es wurde die Rekombination heterologer Polypeptidketten in einem Prozentsatz von 93% beobachtet. Diese „hybriden“ Immunglobulinmoleküle zeigten immunelektrophoretisch die Charakteristica von IgG-Molekülen. Die Ergebnisse bestätigen, daß das experimentelle Vorgehen, das für die Rekombinationsversuche mit mehreren individuellen Myelom-Proteinen bisher benutzt wurde, auch auf radioaktiv markierte Proteine angewendet werden kann.
