Summary
(1) The pattern of insulin secretion after intravenous injection of Ro 6-4563, a new sulphonylurea, is similar to that observed after the intravenous injection of Tolbutamide. Ro 6-4563 may therefore be termed a sulphonylurea “with Tolbutamide-like-action”. (2) A little higher serum insulin levels are observed after the intravenous administration of 25 mg Ro 6-4563 than after administration of 1000 mg Tolbutamide; the changes in the serum concentrations of both glucose and non-esterified fatty acids, however, are almost identical.—With respect to the hypoglycemic effect, 25 mg Ro 6-4563 may be compared to 1000 mg Tolbutamide.
Zusammenfassung
1. Anhand des Sekretionsverhaltens von Insulin ist Ro 6-4563, ein neues Sulfonylharnstoffderivat, in die Reihe der „tolbutamidartig wirkenden Sulfonylharnstoffderivate“ einzuordnen. 2. Die intravenöse Gabe von 25 mg R 6-4563 führt im Vergleich zur intravenösen Gabe von 1000 mg Tolbutamid zu teilweise geringfügig höheren Seruminsulinspiegeln. Die Glucose- und NFS-Werte im Serum sind unter beiden Präparaten jedoch nahezu identisch, so daß 25 mg Ro 6-4563 und 1000 mg Tolbutamid als annähernd gleichwertig zu bezeichnen sind.
