Study of the pancreatic B-cell stimulation in subjects with normal metabolism

Summary

A method is described for maximal B-cell stimulation of the human pancreas. The technique was tested in healthy volunteers. 100 g glucose were given orally 30 minutes before the combined injection of 0.5 g tolbutamide (Hoechst) and 1 mg crystalline glucagon (Lilly). The highest mean immunoreactive insulin concentration obtained was 829 mU/ml (method of Yalow and Berson). This is an appropriate method to measure the secretory capacity of the B-cells after intensive stimulation.

Zusammenfassung

Es wird ein Verfahren zur maximalen B-Zellenstimulation des menschlichen Inselapparates beschrieben, das an Stoffwechselgesunden geprüft wurde. Als Stimulantien dienten Glucose (100 g oral), Tolbutamid (0,5 g i.v.) und Glucagon (1 mg i.v.). Bei kombinierter Anwendung dieser Substanzen wurde eine mittlere maximale Insulin (IMI)-Konzentration von 829 µE/ml Serum erzielt. Das Verfahren ist geeignet, die sekretorische Kapazität der B-Zellen unter maximalen Stimulationsbedingungen zu prüfen.
