Zusammenfassung
Die stabile Sudanophilie der Leukocytengranula beruht ausschließlich auf deren Peroxydaseaktivität. Der Farbstoff Sudanschwarz B dient in Verbindung mit Phenolen und spontan bei der Alterung der Farblösung entstandener Peroxydspuren als Substrat für die endogene Myeloperoxydase. Nach der Zugabe von Meerrettich-Peroxydase zu der Sudanschwarz B-Lösung (nach Sheehen u. Storey) wird in Ausstrichen und Schnittpräparaten die Reaktion in den Zellen der neutrophilen und eosinophilen Granulocytopoese infolge Substrataufbrauchs vollständig gehemmt, während die Anfärbbarkeit echter Lipoide in Fettzellen oder Reticulumzellen des Knochenmarks unbeeinflußt bleibt.
Summary
Stable Sudanophilia of leukocyte granules exclusively depends on their peroxidase activity. In combination with phenols and traces of peroxides, originating in aged staining solutions, Sudan black B may serve as substrate for the endogenous myeloperoxidase. After the addition of horseradish peroxidase to the Sudan black B solution (method of Sheehan and Storey), the reaction in granulocytopoietic cells in smears and tissue sections is completely inhibited due to substrate consumption, whereas the stainability of true lipids in fat cells and reticular cells in bone marrow remains unchanged.
