Zusammenfassung
Die vonEberlein (1960) beschriebene Serumbilirubinfraktionierung wurde einer kritischen Nachprüfung unterzogen. Es konnte festgestellt werden, daß die beim Ausschütteln der Serum-Phosphatpufferschicht mit Butanol und Essigsäureäthylester auftretende Proteinfällung zu teilweise erheblichen Bilirubinverlusten durch Adsorption an den Eiweißniederschlag führt. Die Bilirubinverluste betrugen bei der Butanolextraktion 5–12,8% und bei der Essigsäureäthylesterextraktion 30–36,6% des in den getesteten Albuminlösungen bzw. Seren enthaltenen freien Bilirubins. Die Höhe der Bilirubinverluste erwies sich dabei als abhängig von der Bilirubinkonzentration und dem Albumingehalt. Wegen der hohen Verluste an freiem Bilirubin bei der Essigsäureäthylesterextraktion führt die Errechnung des Monoglucuronidgehaltes als Differenz zwischen der Bilirubinkonzentration in der Butanol- und der Äthylacetatschicht zu fehlerfahten Werten. Die nach dieser Methode gefundenen Ergebnisse können daher nur mit Vorbehalt verwertet werden.
Summary
Eberlein's solvent-partition method for measurement of free and conjugated bilirubin was tested critically by exactly determination of the bilirubin distribution in the three fluid-layers and in the protein precipitation, which develops during the extraction procedure.
It was established, that the extraction of the phosphate buffer-serum mixture by butyl alcohol or ethyl acetate leads to particulary high losses of bilirubin in consequence of absorption on the protein interface.
Butanol and ethyl acetate extraction of solutions of unconjugated bilirubin in albumin or serum by the solvent-partition method was associated with a bilirubin loss of 5–12.8% resp. 30–36.6%. The amount of protein-absorbed bilirubin, which could not be recovered by extraction depended on the bilirubin- and albumin-concentrations of the solutions.
In consequence of the high bilirubin loss during ethyl acetate extraction, the calculation of the bilirubin monoglucuronide value as the difference between bilirubin concentration in the butanol- and the ethyl-acetate layer must give incorrect results.
Hence, the solvent partition method in this form ist not very suitable for accuratly determination of the three fractions of bilirubin in serum.
