Summary
The effects of L-thyroxine and L-triiodothyronine in the ventricular heart muscle were examined. Single papillary heart muscle fibres were stimulated in vitro with a frequency of 1/sec. Membrane resting potential and action potential including rate of rise of the action potential as well as refractory period were measured by means of microelectrodes. A group of guinea pigs was injected daily with L-thyroxine. In another group the acute action of L-triiodothyronine was examined. After acute L-triiodothyronine application as well as after L-thyroxine pretreatment the rate of rise of the action potential was increased. After addition of propranolol this effect was reduced. In hyperthyroid papillary muscles an abbreviation of the refractory period by 35.7 msec (23%) was observed. The findings indicate that thyroid hormones have potential arrhythmic properties in the ventricular myocardium.
Zusammenfassung
Es wurde die Wirkung von Thyroxin und Trijodthyronin auf das Ventrikelmyokard untersucht. Am isolierten Papillarmuskel des Meerschweinchens erfolgten mit intracellulärer Ableitung (Mikroglaselektroden) die Messungen von Ruhe- und Aktionspotential, maximaler Anstiegsgeschwindigkeit und Dauer des Aktionspotentials sowie der Refraktärzeit. Im akuten Versuch wurde L-Trijodthyronin dem Inkubationsmedium zugegeben; im chronischen Versuch wurde 5 Tage lang L-Thyroxin verabreicht. Nach Thyroxin wie nach Trijodthyronin zeigt sich eine signifikante Zunahme der maximalen Anstiegsgeschwindigkeit des Aktionspotentials, die nach Propranolol partiell zurückging. Nach Thyroxinvorbehandlung ergab sich eine Abnahme der Aktionspotentialdauer. Die Refraktärzeit war um 35,7 msec verkürzt. — Thyroxin und Trijodthyronin bewirken bei gleicher Schlagfrequenz am Ventrikelmyokard elektrophysiologische Veränderungen, die die Entstehung von Rhythmusstörungen begünstigen können.
