Rapid demonstration of minor carboanhydrase isoenzyme fractions in human erythrocytes

Summary

A method is described to functionally demonstrate carboanhydrase activity on cellulose-acetate membranes after electrophoresis of human haemolysates. Seven isoenzymes are present in erythrocytes of adults, only five in newborns, and even less in premature infants. There was no abnormal band in an infant with hypothyroidism. A 3 year old boy with juvenile chronic granulocytic leukemia showed the newborn pattern.

Zusammenfassung

Durch Anwendung bekannter histochemischer Methodik ist es möglich, Carboanhydrase funktionell auf Cellulose-Acetat-Folien nachzuweisen. Nach elektrophoretischer Trennung von Hämolysaten finden sich bei Erwachsenen sieben Carboanhydrase-Isoenzymfraktionen, bei Neugeborenen nur fünf und noch weniger bei Frühgeborenen. Bei Hypothyreose trat keine anomale Fraktion auf. Bei einem 3jährigen Jungen mit chronisch myeloischer Leukose vom juvenilen Typ entsprachen die Befunde denen eines Neugeborenen.
