Summary
A radioimmunological method for the determination of plasma renin activity is described using an antibody to angiotensin I. It is shown that this antibody is highly specific for angiotensin 1. During the incubation of plasma at 37° C there was a linear increase in angiotensin I which was neither transformed to angiotensin II nor destroyed by angiotensinases. This radioimmunologic method made it possible to determine plasma renin activity in less than 0.5 ml of plasma, and—under certain experimental conditions—plasma renin concentrations with an excess of exogenous reninsubstrate in as little as 20 µl of plasma.
Zusammenfassung
Es wird eine radioimmunologische Methode zur Bestimmung der Plasma-Renin-Aktivität mit Hilfe eines Antikörpers gegen Angiotensin I beschrieben. Folgende Voraussetzungen waren dabei gewährleistet: 1. der verwendete Antikörper war gegen Angiotensin I spezifisch, 2. das während der Inkubation gebildete Angiotensin I wurde nicht umgewandelt oder abgebaut und 3. der Anstieg des Angiotensin I im Plasma verlief während der Inkubationsdauer linear. Die Methode erlaubt eine Routinebestimmung der Plasma-Renin-Aktivität in weniger als 0.5 ml Plasma und unter bestimmten experimentellen Voraussetzungen, z. B. im Tierversuch, unter Zugabe von exogenem Reninsubstrat die Bestimmung der Plasma-Renin-Konzentration in 20 µl Plasma, so daß bei demselben Versuchstier mehrfache Reninbestimmungen durchgeführt werden können.
