The use of 99m-Tc-Pertechnetate for the determination of red cell volume

Summary

Incubation of human red cells with 99 m-Tc and subsequent addition of SnCl₂ resulted in stable red cell labeling. Simultaneous tagging of the same or another red cell specimen by⁵¹Cr and calculation of the red cell volume from both isotopes gave almost identical results. The 99m-Tc-technique may become the method of choice for red cell volume determination.

Zusammenfassung

Es wurden Untersuchungen zur Bestimmung des Erythrocytenvolumens beim Menschen mit dem kurzlebigen Isotop 99m-Tc durchgeführt. Dabei zeigte sich, daß eine ausreichend stabile Markierung durch halbstündige Inkubation mit 99m-Tc und anschließende kurzzeitige Zugabe von SnCl₂ erreicht werden kann. Die Inkorporationsrate ist vom Plasmaanteil und vom Hämatokrit der zu markierenden Erythrocytensuspension abhängig. Gleichzeitige Markierung derselben oder einer gleichen Probe mit⁵¹Cr ergab gut übereinstimmende Resultate für das Erythrocytenvolumen.
