Zusammenfassung
1. Der Einbau von Acetat in Lipide durch menschliche Lebergewebshomogenatfraktionen wurde untersucht.
2. Optimale Fettsäuresynthese aus Acetat wurde bei einer Mediumzusammensetzung beobachtet, welche der in Tierexperimenten gefundenen ähnlich ist.
3. Einbau von Acetat in Fettsäuren erfolgt nur in Gegenwart sowohl des partikelfreien Überstands als auch von Mikrosomen, steigende Mengen Mikrosomenproteins zeigen einen linearen Anstieg der Fettsäuresynthese aus Acetat.
4. Bei chronisch entzündlichen Lebererkrankungen ist die Fähigkeit des Homogenats, Fettsäuren aus Acetat zu synthetisieren, signifikant vermindert.
Summary
1. The incorporation of acetate into lipids by human liver biopsy homogenate fractions was studied.
2. Cofactor requirements for optimal fatty acid synthesis were found to be similar to those required for fatty acid synthesis from acetate by rat liver homogenate fractions.
3. Acetate incorporation into fatty acids occurred only in the presence of both particle free supernatant and microsomes; increasing amounts of microsomal protein resulted in a linear increase of fatty acid synthesis from acetate.
4. In chronic inflammatory hepatic diseases fatty acid synthesis from acetate in the homogenate is significantly decreased.
