Zusammenfassung
Zur Erweiterung der Kenntnisse über die im menschlichen Körper vorkommenden Enzyme, die das Substrat L-Leucin-β-naphthylamid spalten (LAP), wurden enzymhistochemische Untersuchungen bioptisch gewonnener Dünndarmschleimhaut, quantitative Untersuchungen an Duodenalschleimhaut und Duodenalinhalt vor und nach Cholecystokiningaben durchgeführt sowie die Fraktionierung der LAP mit der Stärkegelelektrophorese vorgenommen. Hohe Enzymaktivität findet sich histochemisch im Zottenepithel der Duodenalschleimhaut. Die elektrophoretische Auftrennung der LAP aus der Schleimhaut ergibt zwei Fraktionen, die sich nach der Wanderungsgeschwindigkeit anders verhalten als die LAP-Fraktionen von Leber- und Pankreasgewebe.
Die Gesamt-LAP-Aktivität im Duodenalsaft ist individuell verschieden und schwankt zwischen 150 und 4600 E. Zu den LAP-Werten des Serums und dem Bilirubingehalt des Duodenalsaftes bestehen keine Beziehungen. Die elektrophoretische Trennung der LAP im Duodenalsaft ergibt regelmäßig zwei starke Fraktionen (Fraktion I und III) und gelegentlich eine dritte schwache Fraktion (Fraktion II). Die am schnellsten wandernde Fraktion (Fraktion I) verändert sich nach Cholecystokiningaben parallel zum Bilirubingehalt und dürfte aus der Galle stammen. Es werden Untersuchungsergebnisse angeführt, die darauf schließen lassen, daß die Fraktion III mit der geringsten Wanderungsgeschwindigkeit ihren Ursprung in der Duodenalschleimhaut hat. Für die Herkunft der Fraktion II aus dem Pankreas ergaben sich Hinweise. Die Art des Übertritts der LAP aus Duodenalschleimhaut ins Duodenallumen sowie mögliche Funktionen des Enzyms in Schleimhaut und Duodenalsaft werden besprochen.
Summary
For better knowledge of enzymes splitting the substrate L-leucyl-β-naphthylamide (LAP) different investigations were performed (enzymehistochemistry of small intestinal mucosa, quantitative determination of enzyme in duodenal mucosa and duodenal juice before and after cholecystokinin medication, and fractionation of LAP by electrophoresis on starch gel medium). High enzyme activity is found histochemically in the epithelium of villi in duodenal mucosa. In electrophoresis of mucosal LAP there are two fractions which differ in their electrophoretic mobility from LAP-fractions of liver and pancreas.
The total LAP-activity in duodenal juice varies from person to person and fluctuates between 150 and 4600 units. It has no correlation to serum-LAP and bilirubin contents of duodenal juice. Generally there are two intensive fractions (I and III) of LAP after electrophoresis and sometimes a third weak fraction (II). After cholecystokinin-medication the fast moving fraction (I) changes corresponding to bilirubin contents and therefore may be a component of bile. Other results show that the slowest fraction (III) originates from the duodenal mucosa and that fraction II may be derived from pancreas. The way of LAP-transport from duodenal mucosa into lumen of duodenum and the possible function of the enzyme in mucosa and duodenal juice are discussed.
