Zusammenfassung
Somatostatin hemmt die Freisetzung einiger Hypophysen-aber auch einiger Intestinal-Hormone und scheint diese Wirkung über eine Hemmung von Adenosin-3′,5′-Monophosphat zu entfalten. Dieser vermutete Antagonismus von Somatostatin und zyklischem AMP wird durch die vorliegenden Untersuchungen bestätigt: Inkubation von Leberschnitten mit zyklischem AMP steigert die Aminosäureinkorporation der aus diesen Schnitten präparierten Ribosomen in vitro mit gleichzeitigem Anstieg des polyribosomalen Anteiles der Leberribosomen. Werden die Leberschnitte zusätzlich zum zyklischen AMP mit Somatostatin inkubiert, so unterbleibt der bei ausschließlicher Inkubation mit zyklischem AMP beobachtete stimulierende Effekt auf die zellfreie Proteinsynthese. Inkubation der Leber mit Somatostatin allein verändert die zellfreie Proteinsynthese gegenüber dem Kontrollversuch nicht. Die Verwendung von Lebergewebe für diese Untersuchungen weist auch darauf hin, daß der mögliche Antagonismus zwischen zyklischem AMP und Somatostatin nicht allein auf endokrines Gewebe beschränkt ist.
Summary
Somatostatin which inhibits the secretion of various pituitary and intestinal hormones has been suspected to exert these effects by inhibiting adenosine-3′,5′-monophosphate accumulation in the respective endocrine gland. Our results, obtained by cell free protein synthesis and by sedimentation through sucrose gradients of ribosomes, prepared from rat liver after incubation with cyclic AMP and/or somatostatin also suggest this antagonism between somatostatin and cyclic AMP. In addition, they indicate that this antagonism is not restricted to endocrine tissues.
