XXIX. Dilatometric micro-estimation of peptidase activity

Summary

A dilatometric micro-method for the estimation of peptidase activity is described. The accuracy of the method is about 50 times that of the previously described micro-technique.

The volume changes following the hydrolysis of alanylglycine were determined and were found to depend onph and the concentration of the phosphate buffer present. The volume changes per millimole of peptide bond split were 9,1–9,9 mm³ in the given cases.

Zusammenfassung

Es wird eine dilatometrische Mikromethode zur Bestimmung der Peptidaseaktivität beschrieben. Die Methode ist ungefähr 50mal genauer als das früher beschriebene Mikroverfahren.

Es wurden die Volumsänderungen bei der Hydrolyse des Alanylglycins bestimmt und festgestellt, daß sie vomph und der Konzentration des vorhandenen Phosphatpuffers abhängen. Die Volumsänderungen je Millimol gespaltener Peptidbindung betrugen in den vorliegenden Fällen 9,1–9,9 mm³.

Les auteurs décrivent une micro-méthode dilatométrique pour déterminer l'activité de la peptidase. Cette méthode est environ cinquante fois plus précise que la micro-méthode décrite auparavant.

Les auteurs déterminèrent les changements de volume après l'hydrolyse de l'alanyleglycine et trouvèrent que ces changements dépendent duph et de la concentration du tampon de phosphate présent. Les changements de volume par molécule milligramme de la liaison-peptide séparée étaient de 9,1–9,9 mm³ dans ces cas.
