Summary
In two cases with plasma cell leukaemia and in one case with a leukaemic variant of M. Waldenström, the leukaemic blast cells of the bone marrow and peripheral blood upon in vivo flash-labelling with3H-TdR, were studied by combined application of cytophotometric determination of the DNA content and by autoradiography.
In plasma cell leukaemia, DNA synthesis time (12–24 h) and generation time (48–60 h) was estimated by observation of the3H-TdR-labelled cells within the cell cycle stages (2c–4c) at different times. The results were confirmed by the data obtained from the labelled mitosis curve.
The leukaemic blast cells showing no labelling one hour after3H-TdR injection were mostly diploid (G0 or G1). The results of the combined cytophotometric and autoradiographic investigations and the labelled mitosis curve allowed for calculation of the size of the G0-compartment indicating that about 46% of the leukaemic blast cells were out of cycle at the time of investigation. The significance of the out-of-cycle cells for treatment and initiation of relapse is discussed.
Zusammenfassung
Bei 2 Fällen von Plasmazelleukämie und einem Fall mit Morbus Waldenström mit leukämischem Blutbild wurden die Blasten im Knochenmark und Blut nach intravenöser Injektion von3H-TdR (flash-Markierung) mit der Kombination von cytophotometrischer DNS-Bestimmung und Autoradiographie untersucht.
Die Beobachtung des Durchganges der3H-TdR markierten Zellen durch die cytophotometrisch ermittelten Zellcyclusstadien (2c–4c) bei den zwei Fällen mit Plasmazelleukämie erlaubte die Ermittlung der DNS-Synthesezeit (12–24 Std) und der Generationszeit (48–60 Std). Diese Daten konnten durch die Ergebnisse der markierten Mitosekurve bestätigt werden.
Die Blasten, die eine Stunde nach3H-TdR-Gabe nicht markiert waren, gehörten fast ausschließlich einer diploiden Zellpopulation an (G0 bzw. G1). Von den cytophotometrisch-autoradiographischen Daten und den Ergebnissen der markierten Mitosekurve wurde bei einem Fall die Größe des G0-Kompartments kalkuliert. Diese macht etwa 46% der Blasten aus. Die Bedeutung der out-of-cycle Zellen für die cytostatische Behandlung und für die Entstehung des Rezidivs wird diskutiert.
