Zusammenfassung
In der beschriebenen Methode benutzten wir die Gel-Permeation als Technik, um Masthilfsmittel von biologischen Substanzen zu isolieren, zu reinigen und zu trennen. Lipidex, ein Hydroxyalkoxypropylderivat von Sephadex mit lipophilen und hydrophoben Eigenschaften, scheint ein ausgezeichnetes Trägermaterial für diesen Zweck zu sein.
Einer ersten Extraktion mit Methanol folgte eine Flüssig-flüssig-Extraktion mit Diäthyläther, und wir haben damit vor der Säulenchromatographie ein ver hältnismäßig klaren Extrakt erhalten. Durch die Verbindung einer „Umkehrchromatographie”, um apolare Lipoide und Cholesterin zu entfernen, und einer „Direktchromatographie”, um eine weitere Reinigung und Trennung durchzuführen, erhielten wir eine östrogene und eine androgene Fraktion. Die beiden Fraktionen wurden dann halbquantitativ durch HPTLC auf Nano-Platten untersucht. Die Induktion einer Fluorescenz auf der Platte durch Zerstäubung einer alkoholischen Lösung von Schwefelsäure ermöglichte den Nachweis.
Summary
In the method described we used the technique of gel permeation to isolate, purify and separate anabolics from biological materials. Lipidex, a hydroxyalkoxypropyl derivative of Sephadex with his lipophilic and hydrophobic properties, seemed to be an excellent support material for this purpose. A primary extraction of the material with methanol followed by a liquid-liquid extraction with diethyl ether, resulted in a relatively clean extract for column chromatography. A combination of a reversed phase chromatography to eliminate apolar lipids and cholesterol and a straight-phase chromatography, to perform further purification and separation, resulted in estrogenic and androgenic fractions. Both fractions were then semi-quantitatively examined by HPTLC on nano-plates. Detection is by induction of a fluorescence on the plate by spraying with an alcoholic solution of sulphuric acid.
