Zusammenfassung
Es wurde ein Reagens, das Titan(III)-chlorid und Bromwasserstoff enthält, entwikelt, welches das Nitrat nach Ansäuerung unter Freisetzung von Stickoxyd in eine Form, wahrscheinlich Nitrit, umwandelt. Auf diese Weise führt die Zugabe des Reagenses bei einem Nahrungsmittel oder einer anderen getrockneten nitrathaltigen Substanz zur Bildung von Stickoxyd, das durch die Anwendung eines Chemoluminescenz-Analysators bestimmt werden kann. Eventuell vorhandenes Nitrit kann von Nitrat durch die Freisetzung von Stickoxyd unter Anwendung von Essigsäure vor der Zugabe des Titan(III)-chlorid-Reagenses unterschieden werden.
Die Variationskoeffizienten für die Stickoxydresultate, die mit 1,0 μg und 10 μg Kaliumnitrat erzielt wurden, waren 12 bzw. 9,2%. Bei einem Niveau von 10 μg KNO3 wurden 97,8±4,2% Stickoxyd, das von einer äquivalenten Menge Nitrit erwartet wurde, aus Nitrat als Folge der Reduktion mit dem Titan(III)-chlorid-Reagens erzielt. Die auf diese Weise durchgeführte Nitratbestimmung wurde durch Ascorbinsäure nicht gestört; es wurde jedoch ein ähnliches Stickoxydresultat mit N-Nitrodimethylamin erzielt.
Summary
A reagent containing titanous chloride and hydrogen bromide has been developed that couverts nitrate to a form, presumably nitrite, releasing nitric oxide on acidification. Thus, thé addition of the reagent to a food or other dried matrix containing nitrate leads to the evolution of nitric oxide, which can be determined using a chemiluminescence analyzer. Any nitrite also present on the matrix cari be differentiated from nitrate by the nitric oxide released using acetic acid prior to the addition of the titanous chloride reagent.
The coefficients of variation for thé nitric oxide responses from 1.0 μg and 10 μg of potassium nitrate were 12 and 9.2%, respectively. At a level of 10 μg of KNO3, 97.8 ±4.2% of nitric oxide expected from an equivalent amount of nitrite was obtained from nitrate, following reduction with the titanous chloride reagent. Ascorbic acid did not interfere with thé détermination of nitrate in this manner, but a similar nitric oxide response was obtained from N-nitrodimethylamine.
