3H-UTP incorporation in polytene chromosomes ofChironomus permeabilized cells

Summary

A method is described that allows the cytological detection of the transcriptional activity of endogenous RNA polymerase in fixedChironomus polytene chromosomes.

3H-UTP is preferentially incorporated onto nucleoli and Balbiani rings where transcription is specially high. The former incorporation was sensitive to actinomycin D while incorporation on Balbiani rings was sensitive to α-amanitin. The label pattern and frequency resemble those detected by3H-uridine incorporation in the living state.
