Summary
InAllium cepa bulb scales the proneness of vacuoles of the upper epidermal cells to asphyxiation,i.e., the fading of neutral red stain under oxygen deficit, decreases from outer to inner scales. Epidermal cells of the second bulb scale show strong asphyxiation while the effect is hardly detectable in the cells of the fourth and the fifth scale.
Upon treatment with 5·10−4 M/l phenylmercuric acetate the fading of neutral red stain from the vacuoles of cells of the second bulb scale is immediate, but is delayed in those of the inner scales; the time lag increases with the position of the scale inwards. At higher concentration (10−3 M/l) the vacuoles of epidermal cells of scales of different ages lose the stain simultaneously. At concentrations weaker than 5·10−10 M/l the loss of stain is caused only through asphyxiation in case it at all occurs.
The results are discussed under the aspect that the ions of neutral red are adsorbed on SH-groups of vacuolar colloids whose concentration decreases with age,i.e., with the position of the scale on the bulb.
Zusammenfassung
Die Bereitschaft zum „Asphyxieeffekt“, d. h. zur Entfärbung unter O2-Abschluß nach vorhergehender Anfärbung mit Neutralrotlösung (pH=7,1) fällt an den Vakuolen der Oberepidermen vonAllium cepa nach innen zu von Zwiebelschuppe zu Zwiebelschuppe ab. Der „Asphyxieeffekt” ist an der zweiten Zwiebelschuppe recht stark, an der vierten und fünften jedoch nicht mehr nachzuweisen.
Die Entfärbung neutralrotgefärbter Vakuolen durch Phenylmercuriacetat in 5 · 10−4 molarer Konzentration läuft an der Epidermis der äußeren (2.) Schuppe rasch ab, erleidet aber an den Epidermen der weiter innen liegenden Schuppen zunehmend eine zeitliche Verzögerung. In höherer Konzentration (10−3 M/l) werden die Epidemien aller unterschiedlich alten Schuppen gleichmäßig rasch entfärbt, bei Vorliegen einer geringeren Konzentration als 5 · 10−4 M/l tritt nur eine Entfärbung durch den „Asphyxieeffekt“ ein (sofern dieser überhaupt eintreten kann), das Gift bleibt weitgehend wirkungslos.
Die Ergebnisse werden unter dem Aspekt diskutiert, daß die Färbung der Vakuole eine Adsorption der Farbstoffionen des Neutralrotes an SH-Gruppen der Vakuolenkolloide ist und daß die Konzentration dieser SH-Gruppen mit steigendem Alter der Zwiebelschuppen abfällt.
