Summary
The water content is very low (10–50%) in dormant plant cells and organs. That is the reason why preparation of those cells can be done—using the freeze-etching technique without any chemical fixation and without incubation of antifreeze medium. So it is possible to show the fine structure of dormant cells without artefacts caused by any pretreatment. After having received the “artefact-free” fine structure of air-dried dormant cells or organs, one can be more successful in showing the ultrastructural changes during passage of dormant cells into the active phase. A simple preparation method is discribed, some examples are demonstrated and the results are discussed with former publications.
Zusammenfassung
Der im allgemeinen geringe Wassergehalt (10–50%) ruhender bzw. lufttrockener pflanzlicher Zellen und Organe ermöglicht eine Präparation mit Hilfe der Gefrierätztechnik, ohne vorherige Fixierung und ohne Verwendung von Gefrierschutzmitteln. Die artefaktfreie Darstellung von Zellen im latenten Lebenszustand ermöglicht erst eine sinnvolle Interpretation der feinstrukturellen Veränderungen, die während des Übergangs ruhender Zellen in die aktive Phase eintreten. Die Methode einer weitgehend artefaktfreien Präparation wird an Beispielen demonstriert und im Zusammenhang mit früheren Ergebnissen diskutiert.
