Biochemical evidence for the histochemical localization of linamarase activity inLotus corniculatus infected withStemphylium loti

Summary

β-glucosidases in healthy trefoil, inStemphylium loti, and inS. loti-infected trefoil have been purified and identified as linamarases byin vitro assays. The location of β-glucosidase activity in these same organisms was determined histochemically by modification of a simultaneous coupling azo-dye technique using bromonaphthyl glucoside as a substrate. The histochemical method was adapted for a quantitative spectrophotometric determination ofin vitro enzyme activity on this substrate. Each purified β-glucosidase was activein vitro on linamarin and the histochemical substrate. With concurrent biochemical and histochemical analyses, it was shown that the localized β-glucosidases were linamarases. Linamarase activityin situ was localized in the phloem, epidermis, and mesophyll in healthy and in infected trefoil leaves and inS. loti hyphae growing either on the surface or in infected tissue.
