Continuous observation of the giant primary nucleus ofAcetabularia in situ

Summary

A technique is described, which allows a continuous microscopical observation of the contents of rhizoids of livingAcetabularia cells. By this technique it was possible for the first time to study all developmental stages of the primary nucleus as well as the growth of the rhizoid. Three types of observation chambers are described, designed for dry and immersion optics and for the use with an inverted microscope.
