Autoradiographic investigation of incorporation of tritiated sucrose and L-methionine in the protonema ofCeratodon purpureus

Summary

The incorporation of sucrose-6,6′-3H (n) and L-methionine (methyl-H 3) into cells ofCeratodon purpureus synthetizing plasmatic filaments in the dark was followed by electron microscopic autoradiography. Sucrose is incorporated almost exclusively into the starch of the chloroplasts but the behaviour of L-methionine is different. With longer exposures to labelled L-methionine (methyl-3H), particularly when they are followed by 3 days in unlabelled medium, most of the silver grains are associated with profiles of endoplasmic reticulum. L-methionine also seems to increase the amount of endoplasmic reticulum in these cells. Only little labelled L-methionine is incorporated into the plasmatic filaments, whereas the coated tubuli are more heavily labelled.
