Summary
The commercial acid phosphatase commonly used in the lead precipitation method for the histochemical demonstration of nuclease, Sigma type I from wheat germ, is itself seriously contaminated with DNase and RNase active at pH 5 and 6. Potato acid phosphatase (Sigma type IV) is free, or nearly free, of DNase and is relatively much less active on RNA. It is as effective as the wheat preparation as a nucleotidase on the two substrates tested, 3′-AMP and 5′-dAMP. Lead nitrate (0.11% concentration) does not fully inhibit the contaminating nuclease of the wheat enzyme.
