Micro determination of α-amino-acids through chelation with copper and titration with EDTA

Summary

A complexometric method was developed for the microdetermination ofα-amino-acids that form soluble copper complexes. The method is based upon reaction with cupric phosphate suspension, isolation of the Cu-complex, its oxidative destruction with conc. nitric acid, evaporation, and titration of the cupric nitrate at pH 7–8 with EDTA in presence of murexide as indicator. Glycine, alanine, valine, tyrosine, serine, histidine-HCl, glutamic acid, lysine-HCl, and aspartic acid give satisfactory results with an average error of ±0.13%.

Zusammenfassung

Eine komplexometrische Methode zur Mikrobestimmung von α-Aminosäuren, die lösliche Kupferkomplexe bilden, wurde ausgearbeitet. Sie beruht auf der Reaktion mit einer Kupfer(II)-phosphatsuspension, der Isolierung des Kupferkomplexes, seiner oxydativen Zerstörung mit Salpetersäure, darauf folgendem Abdampfen und Titration des Kupfernitrats bei pH 7–8 mit ÄDTA in Gegenwart von Murexid als Indikator. Glycin, Alanin, Valin, Tyrosin, Serin, Histidin-HCl, Glutaminsäure, Lysin-HCl und Asparaginsäure ergaben befriedigende Resultate mit einem durchschnittlichen Fehler von ±0,13%.
