The quantitative in situ determination of dinitrophenyl-(DNP-) amino acids by fluorescence quenching on silicagel layers is described. The thin-layer chromatograms on which DNP-amino acids are separated were evaluated with a Turner model 111 fluorometer equipped with a Camag TLC scanner and attached to a 10-mv Heathkit recorder. Parameters such as time, environment and direction of scanning were investigated and optimum conditions determined. The standard deviations for the reproducibility of the technique range from 3 to 9% for 6 amino acids separated in one dimension and from 7 to 13% after twodimensional chromatography. Linear relationships are found between peak areas and logarithm or square root of amino acid concentration. The direct dinitrophenylation of amino acids on the chromatogram prior to development was also investigated and shows some promise.
