Zusammenfassung
Mit Hilfe von Infektiositätsbestimmungen (Pockenzählmethode) und lichtmikroskopischen Studien (Einschlußkörpernachweis, Immunfluoreszenzreaktion) wurde die Vermehrung des Vaccinevirus in HeLa-Zellkulturen unter Einwirkung von p-Fluorphenylalanin (FPA) verfolgt. Durch Zusatz von 100 μg/ml des Antimetaboliten innerhalb des Zeitraumes von 1 Std. vor bis 8 Stdn. nach hochdosierter Infektion läßt sich die Multiplikation des Vaccinevirus statistisch signifikant bis zu 97,5% gegenüber unbehandelten Kontrollen hemmen. Spätere Zugabe bleibt wirkungslos. Die Wirkung ist von 0 bis 6 Stdn. nach Inokulation durch Entfernen des FPA oder kompensierenden Zusatz von Phenylalanin reversibel. Rückwirkungen auf das Virus selbst oder die Inkorporierung in die Zellen konnten ausgeschlossen werden. Das Erscheinen von intraplasmatischen DNA-haltigen Einschlußkörpern und des mit konjugierten Antikörpern erfaßbaren Antigens fällt zeitlich mit dem Anstieg der Infektiosität zusammen. Nach längerer FPA-Einwirkung (12 Stdn. und länger) auf infizierte und normale HeLa-Zellen treten Veränderungen der Zellmorphologie auf, die Störungen des Zellstoffwechsels vermuten lassen. Die Möglichkeiten der FPA-Einwirkung werden besprochen.
Summary
The reproduction of Vaccinia Virus was studied after exposure of HeLa-cells to p-Fluorophenylalanine (FPA) at various periods before and after inoculation by determinations of infectious virus (PFU) and by morphologic studies (inclusion bodies, immunofluorescence). When FPA (100 μg/ml.) was added 1 h before or within 8 h after inoculation with high multiplicity there was no rise of infectivity or increase or detectable viral material inside the cells. FPA, given 12 h and later did not interfere anymore. The effect of the inhibitor was reversible at various stages (0–6 h) either by removing it from the medium or by adding phenylalanine. FPA did not influence the infectious virus or its incorporation into the cells. The formation of inclusion bodies and of viral antigen coincide with the rise of infectivity. After longer exposure to FPA (12 h and more) the morphology of infected and normal HeLa-cells was disturbed. It is suggested that FPA has some deleterious influence on the cell-metabolism. The mechanism of the inhibitory effect of FPA is discussed.
