Zusammenfassung
In Negativkontrast-Präparaten reifer Vaccineviren wurden drei Elementarkörpertypen sichtbar. Das Erscheinungsbild wird durch die Eindringtiefe des Phosphorwolframats bestimmt, die mit steigendem pH-Wert stufenweise zunimmt. Die am besten im Typ 1 dargestellten Filamente gehören der umhüllenden Membran an. Sie werden als Doppelhelix aus Einzelfäden von 25 bis 30 Å Dicke und einer Steigung von etwa 80 Å interpretiert; sie sind DNase-stabil. Am Typ 2 ist zu erkennen, daß die periphere Proteinschicht aus einem Muster von maximal 3500 Untereinheiten besteht. Im Typ 3 gibt sich der DNA-haltige Innenkörper (Core) als ein in Matrix eingebettetes Triplett von Doppelsträngen zu erkennen. Diese bestehen aus etwa 200 Å dicken Einzelsträngen, die um eine axiale Zone von etwa 100 Å gewunden sind und selbst eine Periodizität von etwa 60 Å tragen. Die Interpretation der Virusanatomie beruht auf Abbildung aus allen drei Dimensionen.
Summary
Three different types of elementary bodies are observed in negative stained preparations of mature vaccinia virus. The type of appearance depends on the depth of PTA penetration increasing gradually with rising pH values. The filaments discernible best in type 1 are part of the surrounding membrane. They may represent a double helix of threads 25–30 Å thick with a pitch of about 80 Å. They persist DNase treatment. In type 2 the peripheral protein layer manifests itself as a pattern of about 90 Å periodicity composed of a maximum of 3500 subunits. Type 3 exhibits a matrix-imbedded triplet of double strands at the site of the DNA-containing inner body (core). They consist of single strands about 200 Å thick, having a periodicity of approximately 60 Å, wound up around an axial zone of about 100 Å. The interpretation of the virus anatomy is based on aspects from all three dimensions.
