Summary
The influence of pentagastrin on the gastro-oesophageal sphincter was investigated in healthy mongrel dogs. Pressure measurements were performed applying the withdrawal-perfusion-method of Waldeck (1972). Using a tranquilizing drug or an anaesthesia during surgical operations, it was shown that the real pressure conditions of the lower oesophageal sphincter can not be measured by this procedure. It only registers the resistance value in a narrow segment. The effect of pentagastrin was observed until 3 min after injection and appeared as an increase in tension of the organ. The structural involvement of the muscle fibers of the oesophagus and stomach is the reason why the generally activating effect of pentagastrin on smooth muscle increases the pressure in the sphincteric segment of the oesophagus. The measurement of exact pressure values is reserved to a modified procedure. Some contradictory points in the conception of a hormonal sphincter regulation are discussed.
Zusammenfassung
An Hunden wurde in Combelensedierung und am offenen Thorax in Polamivetintubationsnarkose die Pentagastrinwirkung auf den Speiseröhrenverschluß untersucht. Es zeigte sich, daß die Druckmeßmethode nach Waldeck über die wahren Druckverhältnisse im Verschlußsegment keinen Aufschluß gibt, weil sie nur den Perfusionswiderstand eines engen Segments registriert. Die Pentagastrinwirkung wurde nur bis zu 3 min und als Tonisierung des Oesophagus beobachtet. Die strukturelle Verflechtung von Oesophagas- und Magen-Muskelfasern führt im Rahmen der allgemein tonisierenden Wirkung von Pentagastrin auf glatte Muskulatur des Magen-Darmtrakts zur Erhöhung des Drucks im Verschlußsegment. Die exakte Messung dieses Drucks bleibt einer veränderten Versuchsanordnung vorbehalten. Die Widersprüchlichkeiten in der Vorstellung einer spezifisch hormonellen Verschlußregulation werden diskutiert.
