27. Bestimmung immunologischer Eiweißkörper nach Gewebstransplantationen durch eine simultane qualitativ-quantitative Schnellmethode

Summary

Thermolabile Protein, as occurs after transplantation of organs (liver, kidney) and during rejection crises, cannot be determined by the immuno-radial diffusion method by Mancini, because the antigen will have to be incorporated into the agar at 55°.

A new quick method is therefore presented that has the following advantages:

• (1) It is independent of temperature and at room temperature comprises all protein fractions.

• (2) As it is a simultaneous method it is simple to handle, both qualitatively and quantitatively, in that any number of antisera may be determined on the same plate.

• (3) Qualitative results may be obtained at six hours, quantitative results at 18 hours, in contrast to the 48 hours needed for Mancini's method.

• (4) It is rational and economical, because it may be used for individual sera as well as for several different sera.

The principles and the procedures are described, and the clinical usefullness demonstrated by examples.

Zusammenfassung

Thermolabile Proteine, wie sie nach Organtransplantationen von Leber und Nieren und in der Abstoßungskrise auftreten, sind mit der ImmunoRadialdiffusionsmethode nach Mancini nicht zu erfassen, da das Antigen bei 55° in den Agar inkorporiert werden muß.

Es wird deshalb eine neue Schnellmethode angegeben, die folgende Vorteile hat:

• 1. Sie ist temperaturunabhängig und erfaßt unter Zimmertemperatur alle Eiweißfraktionen.

• 2. Als Simultanmethode ist sie qualitativ und quantitativ einfach zu handhaben, indem beliebig viele Antisera auf ein und derselben Platte bestimmt werden können.

• 3. Eine qualitative Aussage ist bereits nach 6 Std und eine quantitative Analyse nach 18 Std möglich im Gegensatz zu den bisher erforderlichen 48 Std mit der Methode von Mancini.

• 4. Sie ist rationell und ökonomisch, indem sie für einzelne Seren ebenso zu verwenden ist wie für mehrere verschiedene Seren.

  Prinzip und Methode werden dargelegt und die klinische Brauchbarkeit an einschlägigen Beispielen demonstriert.
