Summary
Trasylol® is a basic polypeptide (molecular weight 6,500), IEP at 10.5. It consists of a single peptide chain stabilized by 3 S-S bridges. Trasylol in solution, when at neutrality (below pH 4.0 it detaches itself again), attaches itself reversibly and very quickly to a number of enzymes, e.g., kallikrein, trypsin, chymotrypsin, plasmin, and inhibits their activity. The manifold therapeutic applications of Trasylol are based upon this fact. The s-amino group of the lysine residue in position 15 of the Trasylol molecule is the vital link of the inhibition process. When this group is blocked Trasylol loses its inhibitory action. On i.v. administration Trasylol is stored in the kidney in slightly changed, but still active form. Part of the changed Trasylol is bound to a high-molecular substance, whilst retaining its inhibitory activity.

Trasylol® ist ein basisches Polypeptid (Molekulargewicht 6500), IEP bei 10,5. Es besteht aus einer einzigen Peptidkette, die durch 3 S-SBrücken stabilisiert ist. Trasylol lagert sich in Lösungen um den Neutralpunkt reversibel (unterhalb pH 4,0 wird es wieder frei) an eine Reihe von Enzymen, wie Kallikrein, Trypsin, Chymotrypsin, Plasmin, sehr rasch an, wodurch ihre Aktivität aufgehoben wird. Auf dieser Tatsache beruht die vielseitige therapeutische Anwendbarkeit des Trasylol. Am Hemmvorgang ist die e-Aminogruppe des Lysinrestes in Position 15 des Trasylolmoleküls entscheidend beteiligt. Durch Blockierung dieser Gruppe verliert Trasylol seine Hemmwirkung. Trasylol wird nach i.v. Verabreichung in der Niere in etwas veränderter, aber noch wirksamer Form gespeichert. Unter Beibehaltung seiner inhibitorischen Wirkung ist ein Teil des veränderten Trasylol an eine hochmolekulare Substanz gebunden.
