Zusammenfassung
Eine Gruppe von Imidazolderivaten führt in FL-Zellen bei Konzentrationen von 10 mcg/ml über einen Zeitraum von 8 Stunden zu einer teilweisen, bis etwa 30%igen Reduktion der RNS-Synthese, während die Proteinsynthese mit Ausnahme der Verbindung BC63 unbeeinflußt bleibt.
Nach Infektion mit Mengovirus kommt es zu einer allmählichen Hemmung der zellulären RNS- und Proteinsynthese.
BC59 unterdrückt bei 10 und 20 mcg/ml die virale RNS-Synthese vollständig, auch bei Zugabe 3 Stunden nach t0. Durch eine Applikation 5 Stunden nach t0 kann noch eine Unterbrechung der Virus-RNS-Synthese erreicht werden.
Die durch Virusinfektion ausgelöste allmähliche Hemmung der Proteinsynthese wird durch den Inhibitor wieder zum größten Teil aufgehoben. Es kann angenommen werden, daß wegen Blockade der viralen RNS-Synthese auch keine viralen Hüllproteine gebildet werden.
Summary
Some antiviral derivatives reduce RNA synthesis of FL cells about 30% when given over a period of 8 hours. With one exception host protein synthesis was not influenced.
Mengovirus infected FL cells show a gradual inhibition of cellular RNA and protein synthesis. Viral RNA synthesis increases from 4 hoursp.i.
The compound 1-n-butyl-4-(4′-allyloxyphenyl)-imidazol-hydrochlorid (BC59) applied in concentrations of 10 and 20 μg/ml completely inhibits viral RNA synthesis if administered immediatelyp.i. or 3 hours later. A stop of virus-specific RNA synthesis could be seen even when the agent was given 5 hoursp.i. The gradual inhibition of protein synthesis caused by virus infection can be chiefly reserved by the inhibitor.
