Summary
In 24 melanoma patients body regions with suspicion of metastatic spread were scanned with67Ga-Citrate. Twenty-two out of 28 melanoma metastases (79 %) could be scintigraphically visualized. Falsely negative scans were particularly frequent with small, indurated skin metastases, whereas falsely positive scans were not observed in this study. By means ofin vitro experiments it was shown that the uptake of67Ga into a human melanoma cell line was temperature dependent and reached a maximal value at 37° C after 2 hrs of incubation. Furthermore, it was found that under the same in vitro conditions our melanoma cells bound and released67Ga differently from51Cr. From the release studies it was concluded that67Ga unlike51Cr does not form a stabile intracellular complex with cell proteins.
Zugammenfaggung
Bei 24 Melanompatienten wurden verschiedene metastasenverdächtige Körperregionen mit der67Ga-Szintigraphie dargestellt. Von insgesamt 28 gesicherten Metastasen waren 22 (79 %) szintigraphisch nachweisbar. Falsch negative Befunde wurden besonders bei kleinen derben Hautmetastasen beobachtet, während falsch positive Scans in dieser Studie nicht erhoben wurden.
In einem experimentellen Teil konnte gezeigt werden, daß in vitro die67Ga-Aufnahme in menschliche Melanomzellen temperaturabhängig ist und bei 37° C nach 2 Std Inkubation ein Maximum erreicht hat. Weiterhin wurde gefunden, daß unter gleichen in vitro-Bedingungen Melanomzellen67Ga anders binden und freisetzen als51Cr. Die Releasekinetik von67Ga spricht gegen eine stabile intracelluläre Proteinbindung des Isotops.
