Summary
Dopamine (=3-hydroxytyramine) is the immediate precursor in the biosynthesis of noradrenaline. In brain it probably also serves as a neurotransmitter, particularly in the extrapyramidal system. About 80% of the brain dopamine occur in the neostriatum. The intraneuronal localization of dopamine has been demonstrated by means of histochemical techniques. Reserpine causes dopamine to disappear from the brain and produces a parkinsonian syndrome. The central actions of reserpine can be partially counteracted by increasing the concentration of dopamine in brain (through administration of the precursor L-3,4-dihydrophenylalanine =L-DOPA). In brains of parkinson patients the concentration of dopamine has been found markedly lowered; the parkinson syndrome is favourably influenced by increasing the dopamine concentration (through L-DOPA administration).
Zusammenfassung
Dopamin (=3-Hydroxytyramin) ist die unmittelbare Vorstufe in der Synthese des Noradrenalins. Im Gehirn spielt es wahrscheinlich auch eine Rolle als neurohumoraler Überträger, vor allem im extrapyramidalen System. Etwa 80% des Gehirn-Dopamins ist im Neostriatum lokalisiert. Die intraneuronale Lokalisation des Dopamins ist durch histochemische Untersuchungen wahrscheinlich gemacht worden. Reserpin bewirkt ein Verschwinden des Dopamins und ruft ein Parkinsonsyndrome hervor. Die zentrale Reserpinwirkungen können durch eine Erhöhung der Dopamin-Konzentration im Gehirn (durch Verabreichung der Vorstufe L-3, 4-sonkranker Patienten ist der Dopamin-Gehalt sehr niedrig, und Erhöhung der Dopamin-Konzentration (durch Verabreichung von L-DOPA) beeinflußt das Parkinsonsyndrom in günstiger Richtung.

La dopamine (=3-hydroxytyramine) est le précurseur immédiat dans la biosynthèse de la noradrénaline. Dans le cerveau elle a probablement aussi le rôle d'un neurohumeur, particulièrement dans le systéme extrapyramidal. Environ 80% de la dopamine du cerveau sont localisés dans le néostriatum. La localisations intraneuronale de la dopamine a été démontrée avec une technique histochimique. La réserpine fait disparaître la dopamine du cerveau et provoque le syndrome parkinsonien. Les effets de la réserpine sont supprimés partiellement par l'augmentation de la concentration de dopamine dans le cerveau (après l'administration du précurseur L-3,4-dihydroxyphénylalanine=L-DOPA). Dans des cerveaux de patients souffrant de la maladie de Parkinson on a observé un abaissement marqué de la concentration de dopamine; le syndrome parkinsonien peut être favorablement influencé par l'accumulation de cette amine (après l'administration de L-DOPA).
