Summary
Increased agglutinability by Concanavalin A (ConA) of Ehrlich ascites tumor cells infected with an adapted influenza Ao virus, strain WSA, was maintained despite prefixation with formaldehyde. Noninfected tumor cells, however, were less agglutinable after exposure to the fixative, as has also been reported for transformed cells.
In contrast, the specific binding of125I-ConA was unchanged throughout the infection cycle. Incorporation of viral components into the cell membrane did not provide new sugar moieties accessible to ConA. There was no difference in the number of binding sites between infected and noninfected cells, whether they had been prefixed with formaldehyde or not.
These findings are contrasted with membrane changes observed in transformed cells. As indicated by the experiments with formaldehyde, they reflect stable and irreversible configurational changes of the cell surface and not an altered distribution of ConA receptors by the lectin itself due to differences in membrane fluidity.
