Zusammenfassung
In Kaninchenseren konnten nach Induktion durch Myxoviren zwei verschiedene Interferonkomponenten mittels Gelfiltration über Sephadex-G 100 nachgewiesen werden: Eine hochmolekulare (HM-)Fraktion, die im Säulenausschluß eluiert wurde und somit ein Molgewicht von 100.000 oder mehr besitzt, daneben noch eine niedermolekulare (NM-)Komponente, die als breite Bande eluiert wurde. Ihr Molgewicht liegt etwa im Bereich von 40–50.000.
Bezogen auf die gesamte Interferonaktivität betrug der HM-Anteii nach Induktion durch NDV etwa 20%, nach Induktion durch PR8 dagegen nur 3,5%.
Wurden die Kaninchenseren jedoch in der gleichen Säule mit Puffer + Zusatz von 4m Harnstoff chromatographiert, nachdem die Säulen vorher mit der gleichen Puffer-Harnstoff-Mischung aequilibriert worden waren, so konnte im Ausschluß keine HM-Aktivität mehr nachgewiesen werden. Die NM-Komponente zeigte dagegen stets zwei schwach ausgeprägte Maxima, das eine im Bereich des Molgewichtes 50–55.000, das andere bei 40–43.000.
Experimente an getrennten HM- und NM-Fraktionen ergaben, daß die HM-Komponente offenbar keine Aggregatform von NM-Molekülen ist, sondern wahrscheinlich eine eigene Interferonspezies darstellt. Sie wurde bei Dialyse gegen 2 oder 4m Harnstoff total inaktiviert, während die NM-Komponente keinen nachweisbaren Wirksamkeitsverlust erlitt. Ähnliche Ergebnisse wurden auch nach Dialyse gegen 1 und 2m Guanidin erhalten. Auch in diesem Falle wurde die HM-Aktivität total inaktiviert, während nach der gleichen Behandlung NM-Restaktivitäten von 33–50% übrigblieben.
Schließlich konnte gezeigt werden, daß das HM-Interferon schon nach Hitzebehandlung zwischen 30° und 50° C seine Aktivität verlor. Die NM-Komponente dagegen wurde erst im Bereich von 50–100° C langsam inaktiviert.
Summary
From sera of rabbits induced by myxoviruses two different components showing Interferon activity could be demonstrated by Sephadex-G-100 filtration: a high molecular (HM-)fraction which was eluted in the column exclusion, having therefore a molecular weight of 100.000 or more and a low molecular (NM-)component, being eluted as a broad hand. Its molecular weight lies in the region of 40–50.000.
After induction with NDV 20% of the total interferon activity is composed of the HM-component while after induction with PR8 only 3.5%.
No HM-activity could be detected in the column exclusion in the case the rabbit sera were submitted to chromatography with buffer + 4m urea on the same columns previously equilibrated with the same buffer-urea solution. On the contrary the NM-component always showed two maxima, one in the region of molecular weight 50–55.000, the other at 40–43.000.
Experiments with separated HM- and NM-fractions have shown that the HM-component is not an aggregate of NM-molecules, but probably represents a separate interferon species. It was totally inactivated by dialysis against 2m or 4m urea, whereas the NM-component did not show a demonstrable loss of activity. Similar results were obtained after dialysis against 1 and 2m guanidin. In this case also the HM-activity was totally abolished, whereas after the same treatment 33–50% of the NM-activity was retained.
It could also be shown that the activity of the HM-interferon was lost by heating at 30°–50° C. On the other hand the NM-component was slowly inactivated only at temperatures of 50°–100° C.
