Zusammenfassung
Es wurden die Grana- und Stromalamellen von Tabak- und Sojabohnenchloroplasten nach Fixierung mit Kaliumpermanganat elektronenmikroskopisch untersucht. Die Stromalamellen bestehen aus einer einzigen Lage kugelähnlicher Untereinheiten. Die Granalamellen enthalten zwei Lagen solcher kugelähnlichen Bauelemente, deren Radius etwa 45 Å groß ist. Eine Überschlagsrechnung zur Ermittlung des maximalen Teilchengewichtes und ihr Vergleich mit einem tatsächlich gefundenen Teilchengewicht machen es wahrscheinlich, daß Partikeln solcher Größe in den Lamellen enthalten sein können. Die Untereinheiten enthalten Protein und Lipide. Das Fehlen einer geschlossenen, elektronenmikroskopisch dunklen Randzone und das elektrophoretische Verhalten von Lamellenfragmenten deuten an, daß Proteinanteile bei den Untereinheiten wie auch bei den aus ihnen aufgebauten Lamellen einen Teil der Oberfläche ausmachen. Die Verteilung der Lipide in den Bauelementen wird diskutiert.
Summary
The lamellae within the grana and within the stroma of tobacco chloroplasts and soybean chloroplasts were examinated in the electron microscope after fixation with potassium-permanganate. The stroma lamellae consist of one row of spheroidal subunits. The grana lamellae contain two chains of spheroidal subunits, the radius of which is about 45 Å. A rough estimate of the upper limit of the particle weight compared with a determined one makes it plausible that particles of such a size could exist in the lamellae. The subunits contain protein and lipid; the nonexistance of an unbroken dark rim compared with the electrophoretical behaviour of lamellae fragments indicates the presence of protein within the surface of these subunits and in the superficies of the lamellae. The distribution of lipids in the subunits is discussed.
