IV. Effect of diethylaminoethyl-dextran on adsorption and elution

Summary

Laryngotracheitis virus (LTV), an avian herpesvirus, was adsorbed to chicken kidney cells at 5° C to synchronize penetration. At this temperature virions which adsorbed to cells were sensitive to antibody, and readily eluted in growth medium. Shifting cultures to 37° C caused adsorbed virions to penetrate within a few minutes. Addition of diethylaminoethyl-dextran (DEAE-D) to the inoculum prevented elution at 5° C although adsorbed virions were still susceptible to antibody. The enhanced infectivity produced by DEAE-D in the inoculum was attributed to increased adsorption of virions to cells. When LTV was adsorbed to cells at 5°C and the cells subsequently treated with DEAE-D, an average enhancement of 2.4-fold resulted. This was considerably less enhancement than that produced when DEAE-D is incorporated in the inoculum.
