Zusammenfassung
Es wird über den Einfluß von Cytosin-Arabinosid, Hydroxy-Urea und von Mitomycin C auf die Entstehung membranständiger oder extrahierbarer Antigene, auf die Riesenzellbildung sowie auf die Herpesvirus-bedingte Hemmung der Zell-RNS-Synthese berichtet. Durch die Verabreichung der drei Substanzen wird die Reduplikation der DNS des eingedrungenen Virus völlig blockiert. Trotzdem lassen sich die Phänomene der Riesenzellbildung, der Virus-Antigen-Synthese und der Synthesehemmung der Zell-RNS in vollem Umfange nachweisen.
Des weiteren wurde gefunden, daß UV-Bestrahlung die drei untersuchten Funktionen des Herpesvirus in unterschiedlichem Ausmaß beeinflußt: Nach der Bestrahlung nimmt die Zahl derjenigen Partikel, welche Plaques zu bilden vermögen, stark ab, während die Zahl derjenigen Partikel, die Riesenzellen bilden oder DNS-Synthese anregen, in geringem Maße herabgesetzt wird. Es wird daraus der Schluß gezogen, daß die Fähigkeit zur Induktion der Riesenzellbildung und der DNS-Synthese einen kleinen Treffbereich hat, während die Fähigkeit zur Plaque-Bildung die Mitwirkung von wesentlich mehr Cistrons erfordert.
Summary
The experiments describe the influence of Cytosine-Arabinoside, Hydroxy-Urea and Mitomycin C upon the appearance of herpes-specific membranebound or extractable antigens, upon giant cell formation and the virus induced inhibition of cell-RNA synthesis. The application of these substances inhibits the synthesis of viral-DNA. Under these conditions giant cell formation, antigen-synthesis and inhibition of cell-RNA synthesis are normal (8 hours post infection). Therefore the DNA of the infecting virus-particle is responsible for these virus-induced functions.
UV-irradiation of the infecting virus particles destroys the capacity to form plaques much more readily than the ability to form multinucleated cells or the ability to induce viral DNA-synthesis. The ability to induce giant cell formation or virus DNA-synthesis needs apparently less cistrons than the capacity to form plaques.
