Summary
The authors examinedin vitro the antivirus activity of 5-iodo-2′-deoxyuridine (IDU) against the virus of infectious bovine rhinotracheitis (IBR), in monolayer cultures in liquid nutrient medium and methylcellulose medium, andin vivo in experimentally infected rabbits by cutaneous and corneal scarification.
The nearly complete absence of cytopathogenic effect and the extended period of viral latency in treated cultures, the considerable difference between the virus titers reached in control cultures and those treated with various IDU concentrations, the absence of plaques in monolayer, methylcellulose medium cultures containing 50 γ/ml. of IDU fully prove the inhibitory effect of the substance againstin vitro multiplication of the IBR virus.
From thein vivo tests it must also be concluded that 5-iodo-2′-de-oxyuridine, when locally applied, interferes with development of dermatitic lesions induced by IBR virus in experimentally infected rabbits, on the condition that administration starts immediately after the infection. No toxic effectsin vitro orin vivo were produced by concentrations of 1, 10, 25 and 50γ IDU/ml.
Riassunto
Viene studiata l'attività antivirale della 5-iodo-2′-deossiuridina (IDU) nei confronti del virus della rinotracheite infettiva del bovino (IBR),in vitro, su monostrati cellulari con terreno nutritivo liquido e con terreno alla metilcellulosa,ed in vivo, su conigli infettati sperimentalmente per scarificazione cutanea e corneale.
L'assenza quasi completa dell'effetto citopatico ed il prolungamento del periodo di latenza virale nelle colture trattate, la notevole differenza fra il titolo raggiunto dal virus controllo e quello esposto alle varie concentrazioni di IDU, il mancato sviluppo di placche su monostrati con terreno alla metilcellulosa contenenti 50 γ/ml di IDU, dimostrano chiaramente che detta sostanza è in grado di esplicare un sicuro effetto inibente sulla moltiplicazione del virus IBRin vitro.
Anche le esperienze condottein vivo dimostrano che la 5-iodo-2′-de-ossiuridina, applicata localmente, interferisce sulle lesioni dermiche da virus IBR nei conigli infettati sperimentalmente, purchè l'inizio della somministrazione avvenga subito dopo l'infezione.
Nel corso degli esperimenti non sono stati evidenziati fatti tossici o irritativi locali nei conigli, nè fenomeni citotossici nelle colture cellulari, alle concentrazioni di 1, 10, 25 e 50 γ di IDU/ml.
