Zusammenfassung
Die Infektiosität und die virusspezifische antigene Aktivität hochgereinigter Poliovirus-Konzentrate konnten durch Behandlung mit salpetriger Säure unter verschiedenen Bedingungen (0,1-m. NaNO2/pH 4,6/ 20° C/12h und 0,5-m. NaNO2/pH 4,0/20° C/2 h) entweder vollständig, oder aber weitgehend zerstört werden, ohne daß dabei ein Zerfall der Virusteilchen oder sonstige in der analytischen Ultrazentrifuge erfaßbare Veränderungen der Partikelstruktur erfolgten. Erst bei länger dauernder Einwirkung der salpetrigen Säure unter den Bedingungen 0,5-m. NaNO2/ pH 4,0 wurde ein Zerfall von Virusteilchen festgestellt. Als Zerfallsprodukte konnten eine vermutlich aus leeren Proteinhüllen bestehende Komponente mit s20 ∼ 90 S und freie, jedoch stark degradierte Virusnucleinsäure nachgewiesen werden.
Summary
The infectivity and the specific antigenic activity of highly purified poliovirus concentrates could be destroyed completely or to a considerable extent by nitrous acid under different conditions (0.1 M NaNO2/pH 4.6/20° C/ 12 h and 0.5 M NaNO2/pH 4.0/20° C/2 h), but without an indication by analytical ultracentrifugation results of any disintegration or other structural change of the virus particles. When 0.5 M nitrous acid was acting at pH 4.0 for longer times, however, there were indications of particle disintegration. Disintegration products were a component with s20∼ 90 S, probably representing empty capsids, and free but strongly degraded virus nucleic acid.
