Zusammenfassung
Die thermische Inaktivierung von Poliomyelitisviren Typ I (Stamm Mahoney), die in Gewebekulturen trypsinaufgeschlossener Affennieren vermehrt worden waren, wurde im Temperaturbereich um +45°C untersucht. Es ergab sich, daß in den Virussuspensionen enthaltene niedermolekulare Substanzen die thermische Inaktivierung stark beschleunigen können. Diese Substanzen konnten durch Dialysen entfernt werden, wodurch die thermische Resistenz erheblich verstärkt wurde. Es gelang außerdem durch Komplexbildner wie EDTA und, wenn auch in schwächerem Maß, KCN, die beschleunigende Wirkung auf die thermische Inaktivierung aufzuhalten. Der Zusatz von Aminosäuren hatte einen analogen Effekt. Da diese Befunde auf die mögliche Beteiligung von Metallionen hinwiesen, wurde eine Reihe von Salzen untersucht. Dabei wurde gefunden, daß Salze des dreiwertigen Chroms eine stark beschleunigende Wirkung auf die thermische Inaktivierung besitzen.
Summary
Studies on the thermal inactivation of poliomyelitis virus type 1 (Mahoney), prepared in monolayer tissue cultures of trypsinized monkey kidney cells, were made at a temperature of 45° C. Evidence was furnished of the presence in the virus fluid of substances of low molecular weight, which increase the rate of thermal inactivation. These substances could be eliminated by dialysis, whereby thermal resistance became greatly increased. Inhibition of thermal inactivation was possible by addition of chelating agents as EDTA and partly by KCN. Addition of aminoacids had a similar effect. The data pointed to the possible role of metal ions. Therefore we tested the effect of some metal salts. Cr+3 — salts increased the rate of thermal inactivation.
