Zusammenfassung
Eine Methode zur Bestimmung der Wanderungsgeschwindigkeit (Beweglichkeitskonstanten u) von optisch nicht erfaßbaren Substanzen, wie Viren oder anderen aktiven Stoffen, in der trägerfreien Zonenelektrophorese in einem Glukose-Dichtegradienten wird beschrieben. Gegenüber den bisher gebräuchlichen Verfahren erfolgt das Fraktionieren während der Elektrophorese. Gemessen wird die Wanderungszeit (Medianzeit 50%) der Substanz über eine konstante Wegstrecke. Der Glukose-Dichtegradient gewährleistet aufgrund der Konstanz des Produktes: Leitfähigkeit · Viskosität die gleichbleibende Wanderungsgeschwindigkeit über die gesamte Weglänge.
Ausführlich wird auf die Theorie der trägerfreien Zonenelektrophorese in einem Dichtegradienten eingegangen und dabei eine neue, formunabhängige Stoffkonstante e/A oder formabhängige e/r (e = Ladung und A = Formfaktor eines wandernden Teilchens — bei sphärischen Teilchen = dem Radius r) als zusätzliche charakteristische Konstante zur Diskussion gestellt. Am Beispiel eines Elektrophoreseversuches mit reinem Rinderalbumin werden die theoretischen Betrachtungen bestätigt.
Summary
A method is described using the carrierfree zone electrophoresis with a density gradient of glucose. This technique is useful for the determination of the migration velocity (electrophoretic mobility u) of substances as viruses or other active materials non-detectable by optical systems. Contrary to the methods used so far the fractionation takes place during electrophoresis. The migration time (median time 50%) of the substance is measured over a constant distance. The density gradient of glucose guaranteed the uniform migration velocity over the whole path because of the constant of the product: conductivity · viscosity.
The theory of the carrierfree zone electrophoresis with a density gradient is discussed in detail. Also a new substance constant independent e/A or dependent e/r on its form (e = charge and A = formfactor of a migratory particle — in case of spherical particles = radius r) is discussed as an additional characteristics constant. Using pure albumin of cattle as an example of an electrophoresis the theoretical considerations are confirmed.
