Zusammenfassung
Sowohl bei der Positiv-wie bei der Negativkontrastierung von Reovirus Typ 3 kann das Auftreten sogenannter „voller“ und „leerer“ Formen durch Wahl geeigneter Präparationsbedingungen gesteuert werden. An der Projektionsperipherie beider Formen ist abzulesen, daß das äußere Capsid aus 92 Capsomeren besteht, die feine Achsen von etwa 15 Å besitzen und in einem Abstand von etwa 35 Å angeordnet sind.
Im inneren Bereich der Projektion von „vollen“ und Übergangsformen kommt es zur Darstelhng des inneren Capsids, besonders deutlich bei Abbildung in Richtung der fünfzähligen Symmetrieachse. Das innere Capsid besteht aus 42 Untereinheiten mit Achsen von 40 bis 50 Å Durch-messer und einer Wandstärke von etwa 25 Å. Die Symmetrieachsen beider Capside decken sich miteinander.
Unter geeigneten Bedingungen gelingt reproduzierbar die Absprengung und Zerlegung des äußeren Capsids in Capsomeren. Die Abbildung des dabei freigelegten Innenkörpers entspricht der des im intakten Viruspartikel sichtbar werdenden inneren Capsids.
Summary
The amount of the so-called “full” and “empty” particles in specimens of reovirus type 3 can be varied at will both in positive and negative staining.
From the periphery of the particles it can be deduced that the outer capsid consists of 92 capsomeres exhibiting an axis of about 15 Å and being separated by a distance of about 35 Å.
The inner part of the projection of “full” and intermediate particles represents the structure of the inner capsid particulary well defined in virions standing on the axis of 5-fold symmetry. The inner capsid is built up of 42 morphological units with axes of 40 to 50 Å in diameter and walls of about 25 Å thickness. The axes of symmetry of both capsids are identical.
Under suitable conditions the outer capsid may be decomposed into the capsomeres. The appearance of the inner body thus liberated corresponds to the inner capsid visible in complete particles.
